New thiosemicarbazones I [R1 = H, 3-Cl, 4-F, etc.; R2 = H, OMe] were synthesized via condensation of N-benzylindole-3-carboxaldehydes with N4-substituted thiosemicarbazides in excellent yield. These thiosemicarbazones I were reacted with Et bromoacetate to produce original heterocyclic-substituted indole derivatives possessing a 4-oxo-thiazolidine group II. Anal. IR and NMR spectra and elemental anal. were performed to reveal their structures. The antimicrobial activity of all synthesized compounds was evaluated for antibacterial activity in vitro against Gram-pos. and Gram-neg. bacteria. Antibacterial screening data showed that two compounds I and II demonstrated activity against Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa. These preliminary results indicated that newly synthesized compounds such as I [R1 = 3-Cl, R2 = OMe] and II [R1 = H, R2 = H] showed a promising antibacterial potency.

The applications of Cu(II)-β-CD as water soluble green catalyst for the one-pot conversion benzyl halide to benzyl azide and 1,2,3-trizoles at room temperature and very short reaction time (15-70 min) was presented. Products were separated without any workup, simple by filtration, with >99 % selectivity and purity. As-synthesized material was characterized by, HR-TEM, XPS, FESEM, FT-IR, TGA and XRD to understand its structural properties as well as attachment of copper by host-guest interaction. This catalyst also showed effectiveness for the condensed ketone and aldehydes moiety, which produced biofuels intermediate. The recyclability of this catalytic system was tested up to five cycles and only a very slight loss of catalytic activity was detected.

Cathepsin B is a lysosomal cysteine protease that has various physiol. and pathophysiol. functions. We present here the discovery of 6-substituted 4-benzylthio-1,3,5-triazin-2(1H)-ones as inhibitors of cathepsin B, starting from screening of a library of variously 2,4,6-trisubstituted 1,3,5-triazines and 1,3,5-triazin-2(1H)-ones on three different human cathepsins. The synthesis and enzymic evaluation of a focused library of new 1,3,5-triazin-2(1H)-ones, e.g. I [R – o-OMe, m-F, p-NO2, etc.], is also described. The detailed kinetics analyses have shown that these compounds can act as reversible, partial mixed-type inhibitors of cathepsin B, with Ki and Ki‘ values in the low micromolar range. The inhibitory activities of selected compounds were also assessed against two related cysteine proteases, cathepsin H and cathepsin L, to estimate their selectivity; these compounds have a selective profile for catB and catL over catH.

For more in-depth exploration of the chem. space around the entrance channel of HIV-1 reverse transcriptase (RT), a series of novel indolylarylsulfones (IASs) bearing different chiral N-substituted pyrrolidines (R/S)-I [R1 = methanesulfonyl, pyridin-3-ylmethyl, (2-fluorophenyl)methyl, (4-methyl-5-oxo-4,5-dihydro-1H-1,2,4-triazol-3-yl)methyl, etc.], azetidines II or substituted sulfonamide groups III [R2 = 2-(2,4-dioxo-1,2,3,4-tetrahydropyrimidin-1-yl)ethyl, ethanesulfonyl, cyclopropanesulfonyl, etc.] at indole-2-carboxamide were designed and synthesized as potent HIV NNRTIs by structure-guided scaffold morphing approach. All the IASs exhibited moderate to excellent potency against wild-type HIV-1 with EC50values ranging from 0.0043 μM to 4.42 μM. Notably, compound (R)-I (R1 = methanesulfonyl) (EC50 = 4.7 nM, SI = 5183) and (S)-I (R1 = methanesulfonyl) (EC50 = 4.3 nM, SI = 7083) were identified as the most potent compounds, which were more active than nevirapine, lamivudine and efavirenz, and also reached the same order of etravirine. Furthermore, some compounds maintained excellent activity against various single HIV-1 mutants (L100I, K103 N, E138K, Y181C) as well as one double mutant (F227L/V106A) with EC50 values in low-micromolar concentration ranges. Notably, II (R2 = carbamoylmethyl) displayed outstanding potency against F227L/V106A (EC50 = 0.094 μM), and also showed exceptional activity against E138K (EC50 = 0.014 μM), L100I (EC50 = 0.011 μM) and K103 N (EC50 = 0.025 μM). Addnl., most compounds showed markedly reduced cytotoxicity (CC50) compared to lead compounds, especially II [R2 = (2-cyanophenyl)methyl] (CC50 >234.91 μM, SI >18727) and II (R2 = methanesulfonyl) (CC50 >252.49 μM, SI >15152). Preliminary SARs and mol. modeling studies were also discussed in detail, which may provide valuable insights for further optimization.

The paper aimed to study a new synthetic method of aromatic acyl cyanides. Catalyzed by samarium triflate [Sm(OTf)3] aromatic acyl chlorides was reacted with sodium cyanide to give aromatic acyl cyanides. Under the optimal reaction conditions, the yields and purities of aromatic acyl products reached about 90% and above 98% resp. It was a novel practical method to prepare aromatic acyl cyanides, with mild reaction conditions and high selectivity, and without the use of phase transfer catalyst.

A series of novel triazolinones were synthesized and their structures were characterized by 1H NMR, elemental anal. and single-crystal X-ray diffraction anal. The herbicidal activities were evaluated against Echinochloa crusgalli (L.) Beauv., Digitaria adscendens, Brassica napus and Amaranthus retroflexus. The herbicidal activity data indicated that the title compounds had higher activities with substituted benzyl group moieties than with other groups such as sulfonyl, alkyl, etc. To further investigate the structure-activity relationship, comparative mol. field anal. was performed on the basis of herbicidal activity data. Both the steric and electronic field distributions of comparative mol. field anal. are in good agreement in this work. The results showed that a bulky and electroneg. group around the ortho- or para-positions of the benzene ring would possibly lead to higher activity. Based on the comparative mol. field anal., compound 2-[(1-(2,4-Dichlorophenyl)-3-methyl-5-oxo-1H-1,2,4-triazol-4(5H)-yl)methyl]benzonitrile was designed and synthesized, which displays as good herbicidal activities as the com. herbicide, carfentrazone-Et. The activity against Digitaria adscendens is 66.1% under preemergence at 300 g of a.i./ha.

A series of copper(II)-Schiff bases-functionalized polyacrylonitrile fiber catalysts was successfully prepared using copper acetate as copper source and characterized by elemental anal., Fourier-transfer IR spectroscopy, UV-visible spectroscopy, XPS and inductively coupled plasma anal. Excellent phys. strength and thermal stability of the fiber catalysts were demonstrated by SEM, X-ray diffraction, thermogravimetric/differential scanning calorimetry anal. and mech. strength measurements. Furthermore, these catalysts were successfully applied to one-pot multicomponent copper-catalyzed azide-alkyne cycloaddition (CuAAC) reaction and aldehyde, alkyne, amine (A3) coupling reaction in which the influence of different substituent groups on the catalytic activities of fiber catalysts were investigated in detail. Among them, the bis[N-ethyl-3,5-di-tert-butyl-salicylideneiminato]copper(II)-functionalized polyacrylonitrile fiber (PANS2F-Cu) as a green, efficient catalyst exhibited the best catalytic activity as its high hydrophobic micro-environment could aggregate the reactants to the catalytic sites and accelerate the reaction. In addition, the PANS2F-Cu performed well in scaled-up experiment and showed excellent recyclability (at least ten times), and these characteristics enabled it to have great potential for further applications.
