R2R3NCH:CR1CR:CXCN [R,R1 = alkyl, aryl; RR1 = (CH2)n (n = 3-5); R2, R3 = alkyl, aryl; R2R3 = (CH2)4, (CH2)5, (CH2)2O(CH)2; X = CN, carbonyl such as acyl, alkoxycarbonyl, H2NCO, NO2, aryl], useful as synthons for pharmaceuticals were prepared by reaction of R1CH2CR:CXCN with HC(OR4)3 (R4 = alkyl) and HNR2R3 in the presence of acidic condensation agent by 8 methods. A mixture of 2-O2NC6H4CO2H and 0.01 mol HNPhMe was treated with 0.015 mol HC(OEt)3 and 0.01 mol Me2C:C(CN)2 and the mixture refluxed 30 min. to give 65% MePhNCH:CHCMe:C(CN)2.
